No. It does not. Once you have a mark in an achivement it will never go away unless otherwise stated. (Ex. In one AB assault 3 nodes, in 15 seconds, kill 50 dragon whelps, etc)
This the hardest achievement? Doubt it, one mask isn't that hard at all, all 20 masks; now that's hard!
