The totem seems to have about a 1 minute cooldown and only one boss can be spawned in the arena at any time. I went up to the arena with all 3 totems and found that I got the "you can't do that yet" error when attempting to use the next totem for some time after the last boss died (usually about as long as it took the boss's body to disappear + ~10 secs).
All three quest mobs are open-tap, meaning you can just wait around for others to summon them as well.
