While you can free the captive while mounted this does not currently dismiss the rune trap. Because the rune trap does not disappear the captive is not respawning. I tested this on a few runes and by dismissing the rune trap it forced the captive respawn in about a minute. Unfortunately you have to dismount to dismiss the rune. This has been my experience and could use some more corroboration.

I have to laugh at how angry the Magister gets when they've got Kirin Tor captives stuck around their base too.
The runes are in the Violet Rise area on the Isle of Thunder ~ Coordinates 60, 70
Runes are blue circles on the ground and you need to click on them.
Empty runes will not count towards the quest, the runes must contain a captive to count. Its quite annoying as there sometimes are no captives at all and you have to run through the whole alliance site!

The Kirin Tor have gone too far! Now they hold our fellow Sunreavers as war prisoners! There will be no discussion of prisoner exchange. We will handle this ourselves. Go to Violet Rise and free as many prisoners as you can. Do not get captured yourself. We cannot afford to lose you too.
