Before you engage [npc=160143], you’ll want to pet [npc=171148], located at the small hill at 63.2, 30.8.

The [npc=160143] starts in a cave, with the entrance being 62.2, 28.9. After damaging her a bit, she burrows to another cave, entrance 61.4, 33.4. Only after that does she show up in the "Matriarch's Den", where [npc=160155] is waiting. Don't make the mistake of waiting in that chamber for her to spawn, because she never will until you fight her in the other two caves.
Ultimately, the quest objectives on the map will lead you to where you need to go!

You can use the [item=173534] ability on the [npc=160143], and it will stun her like it does for the other gorms. Helps to neutralize most of the things she does.

Side note - if you are running around this general area and wondering why you keep getting dismounted, it is because the [npc=170227]e burst when run over. The burst does damage, and is apparently capable of dazing you.
EDIT: After doing this quest and its world quest equivalent numerous times now on live servers, it seems the damage from the larva burst has been removed.

Description

We must go after Master Sha'lor. Tirna Vaal needs him! And I... I... I will report to the droman. They must know that Sha'lor is in danger. Then, I will enter the burrows myself from the south and help you defeat it. For now, see what you can find. I refuse to believe that he is gone. That monster will know the fury of a vorkai.

Pins are the quest’s own POI points from the current retail client, projected onto the in-game zone map. The game marks one blob as the quest’s turn-in; the rest are objective areas. Points that fall outside a map’s window are dropped rather than guessed at.

Where this quest fits

Before this quest

Taken from the storyline's own step order.

  • Finish What He StartedArdenweald · Level 10-56

Finding Sha'lor is our main priority. But there is another opportunity. Take the Gormherd Branch that he dropped. I know he was tinkering with it so that it would work on wild gorm. Use it on the gorm in the burrows, and see if you can collect any of their gorm gris.

After this quest

Taken from the storyline's own step order.

  • Burrows AwayArdenweald · Level 10-56

<The ground has been freshly dug, and there are signs of more tunnels all over the place. Collapsing a few burrows would go a long way toward keeping Tirna Vaal safe from gorm infestation.>
